Chloramphenicol is een breed spectrum antibioticum.Target: AntibacterialChloramfenicol is een bacteriestatisch geneesmiddel dat de bacteriële groei tegenhoudt door de eiwitsynthese te remmen. Chloramphenicol voorkomt verlenging van de eiwitketen door de peptidyll-transferase-activiteit van het bacterieel ribosoom te remmen. Het bindt specifiek aan residu's van de A2451 en A2452 in de 23S rRNA van de RIBOSOMALE subeenheid VAN DE JAREN VIJFTIG, waardoor peptide-bindingvorming wordt voorkomen. Terwijl chloramphenicol en de macrolide klasse van antibiotica allebei met ribosomes interactie aangaan, is chloramphenicol geen macrolide. Het verstoort direct substraatbinding, terwijl macroliden de progressie van het groeiende peptide sterisch blokkeren.
Toepassing&functie

Vanwege weerstand en veiligheidsoverwegingen is het niet langer een eerstelijns middel voor enige indicatie in ontwikkelde landen,
hoewel het soms topisch wordt gebruikt voor ooginfecties, heeft het mondiale probleem van het bevorderen van bacteriële resistentie tegen nieuwere medicijnen geleid tot hernieuwde belangstelling voor het gebruik ervan. In landen met lage inkomens wordt chloramphenicol nog steeds op grote schaal gebruikt omdat het buitengewoon goedkoop is en gemakkelijk beschikbaar is.
